Bachem Webinar – Superior MS-compatible analytical method for determining oligonucleotide peak purity

Bubendorf, Switzerland: – The latest webinar from TIDES manufacturer Bachem will describe a novel two-dimensional modelling approach to determining oligonucleotide purity in liquid chromatography (LC) using strong anion exchange (SAX) methods that makes it suitable for Mass Spectrometry (MS) analysis.

The hour-long webinar ‘2D LC/MS: Peak purity determination for denaturing SAX oligonucleotide methods’ is being hosted on the Zoom platform on May 20, 2026, commencing 1700 hrs. CET (UT +2) and is free to subscribe (see Resources). It will be of great interest to any researcher looking for superior analytical methods to determine the purity and related impurities of oligomers as emerging new therapeutic entities.

Overcoming the MS barrier 

SAX LC is often used as an orthogonal approach to determining oligo purity, in conjunction with well-established ion-pairing reversed phase (IP-RP) methods, particularly for phosphorothioate oligonucleotides. However, a recurrent problem with these approaches is that the non-volatile eluents used in SAX LS make them incompatible with the MS analysis needed for direct peak purity analysis.

The Bachem webinar will describe a novel 2D-LC-MS approach it has developed in-house that allows the evaluation of peak purity by using SAX methods in combination with IP-RP and high-resolution MS.

The discussion will be led by two Bachem scientists with in-depth knowledge of oligonucleotide analysis.

• Marion Fresch is senior scientist in the QC Early Phase department at Bachem AG. Since joining Bachem five years ago, she has specialized in HPLC method development, validations, forced degradation studies, release analytics, and stability testing for oligonucleotide NCE projects.
• Claire Meyer holds aa PhD in Chemistry from the University of Basel and completed her postdoctoral research in Bioengineering at the University of California. She began her industry career as a Proposal Manager in a Biologics CDMO, before joining Bachem, where she has focused on oligonucleotides, transitioning from proposal management into project management. Her expertise spans early-phase CMC for oligos, while driving continuous improvement initiatives.
