Leading CDMO Bachem has released an updated version of its oligonucleotide therapeutics brochure.

The technical paper explains how oligo-based therapeutics are redefining what is possible in targeted therapies by expanding the druggable genome, and enabling precision treatments across a growing range of diseases.

Gearing up for NextGen Therapies

The downloadable brochure also shows the importance of innovative synthesis methods in revolutionizing the large-scale production of the oligonucleotides needed to power these NextGen therapies.

The paper traces how Bachem supports its partners across the full oligonucleotide lifecycle, from early development to commercial-scale manufacturing, combining deep expertise across various modalities (including siRNA, ASO and conjugates) with advanced technologies and scalable production capabilities.

New Solid and Liquid Phase approaches

This integrated approach, from synthesis and chemical modifications through purification and analytical support, ensures high-quality, reliable delivery at every stage of the project. Innovative approaches such as fragment synthesis and enzymatic ligation can overcome the challenges of using traditional peptide chemistry to produce longer, chemically complex oligomer sequences  at required yield, scalability, and environmental sustainability.

The bulletin explores the role of fragment synthesis with other innovations such as Stirred Bed Technology (SBT SPOS), and Tag-Assisted One-Pot Liquid Phase Oligonucleotide Synthesis (TOP LPOS) in enabling large-scale, sustainable manufacturing. It shows how these can be applied across a range of common applications, including siRNA, oligonucleotide conjugates, and complex molecular structures.

Efficiency and Sustainability

Specifically, the white paper explains how enzymatic ligation can improve cost efficiency, reduce chemical waste, and support greener production processes, with Bachem’s enzymatic ligation processes now routinely achieving >90–95% purity and higher overall yields.
