Presenting a continuous plasmid DNA vision at ISPPP 2025

Vienna, Austria: – enGenes Biotech GmbH (enGenes) will share details of its progress towards the world’s first platform for truly continuous production of plasmid DNA (pDNA) with the bioscience community at the 44th International Symposium on the Separation of Proteins, Peptides and Polynucleotides (ISPPP) in Munich.

As part of the Continuous and Intensified Processing session, enGenes Senior Bioprocess Scientist Dr. Markus Berg will present “Continuous Downstream Processing for Plasmid DNA: Advances in Integrated Separation and Purification.” (November 10th at 16:50)

Urgent need

Dr. Berg will provide background on the formation of the iCAP (Integrated Continuous and Automated Platform for Plasmid Production) project to address the growing need for more efficient and scalable separation of plasmid DNA, a critical step in producing gene therapy vectors, mRNA vaccines, and DNA-based therapeutics.

The project reunites enGenes with established partners Novasign GmbH and Qubicon AG, having previously collaborated on the EConti project for continuous protein production.

Integrating upstream and downstream

Dr. Berg will show how the iCAP project is developing a fully continuous and automated pilot system integrating upstream fermentation with continuous downstream purification. The latter combines continuous alkaline lysis with continuous chromatography to enable uninterrupted recovery and purification of pDNA with high yield and quality.

He will also highlight the importance of IT, with real-time monitoring and model-predictive control implemented via a digital twin framework, enhancing process stability and robustness. The presentation will also underline enGenes’ specialist expertise in genetically stabilized E. coli strains to support sustained production under continuous conditions.

“I hope to show the ISPPP community how iCAP represents a potential revolution,” said Dr. Berg.

“If successful, we can create a stable integrated platform that significantly reduces pDNA production costs, improves process sustainability, and establishes new standards for continuous polynucleotide separation at industrial scale,” he explained.

About ISPPP 2025

The International Symposium on the Separation of Proteins, Peptides and Polynucleotides (ISPPP) is an annual conference for scientists from around the world to share knowledge on the separation, purification, and characterization of biologically important molecules.

It focuses on key developments and novel approaches to the productivity, regulatory and financial challenges of biological molecule separations, purification, and analysis.

Founded in 1981, the symposium has latterly been held in alternate year locations in Europe and the USA, with the previous meeting held in Florida.

The 44th International Symposium on the Separation of Proteins, Peptides and Polynucleotides (ISPPP XLIV) will be a four-day event opening November 9, 2025, at the Garching Campus of Technical University Munich, featuring a three-day plenary program preceded by a workshop day.
