Evercyte derived cell lines
Evercyte has developed cell lines from adipose tissue and bone marrow-derived mesenchymal stem cells (MSCs) that can be grown without limitations, show typical MSC markers and functions and therefore represent valuable models for studying processes such as differentiation, inflammation, tissue homeostasis and repair.
This is in line with increasing evidence that the potency of MSCs is at least partly mediated by secreted extracellular vesicles (EVs).
Both the ASC/TERT300 adipose-derived and the BM-MSC/TERT292 bone marrow-derived MSC cell lines have been established under conditions that will allow cGMP production of EVs for clinical applications.
Adipose derived MSCs
The ASC/TERT300 cell line (Catalog number: CHT-001-0300) has been developed from human adipose-derived mesenchymal stem cells showing typical markers and functions of MSCs to provide a valuable model for studying processes such as differentiation, inflammation, tissue homeostasis and repair.
The cell line exhibits mesenchymal, spindle-shaped morphology and has a life span extension based on ectopic expression of human telomerase reverse transcriptase (hTERT).
ASC/TERT300 cells can be induced to differentiate towards adipocytes (oil red O staining), osteoblasts (Alizarin red staining) and chondrocytes (Alcian blue staining).
Compared to the earlier ASC/TERT1 cell line that was used exclusively for scientific purposes, ASC/TERT300 is manufactured under conditions that will allow later use for production of GMP grade extracellular vesicles.
Bone marrow derived MSCs
Evercyte ́s BM-MSC/TERT292 cell line (Catalog number: CHT-063-0292) has been developed from bone marrow-derived mesenchymal stem cells, can be grown without limitations, shows typical markers and functions of MSCs and therefore represents a valuable model for studying processes such as differentiation, inflammation, tissue homeostasis and repair.
This cell line also exhibits mesenchymal, spindle-shaped morphology and has a life span extension based on ectopic expression of human telomerase reverse transcriptase (hTERT).
Ready for cGMP production
The ASC/TERT300 and BM-MSC/TERT292 cell lines have both been established under xeno-free conditions with full documentation of all manipulation steps. Both Evercyte cell banks can be transferred to a GMP facility for establishment of a production master cell bank.
This speeds the way to cGMP production of EVs for clinical applications at very high quality, which includes being free from bacterial contaminations (including mycoplasma, fungi, HIV, HAV, HBV, HCV, Parvo-B19) and cross-contaminations.
Future derived cell lines
Evercyte is now developing placenta-derived MSC stem cell lines than can also be transferred to cGMP production purposes.
These new lines will be derived from human amniotic membrane and placental tissue cells.
