With more than 100 years experience in phosgene chemistry, ISOCHEM is one of the few manufacturers in the world that can produce challenging phosgene derivatives at scales ranging from kilograms to full-scale production in the hundreds of tonnes.

In total, ISOCHEM offers more than 120 phosgene-derived products. It can manufacture phosgene derivates at pilot or full commercial sale and to GMP or non GMP standards according to requirements.

ISOCHEM can supply conventional phosgene derivatives including chloroformates, acid chlorides, isocyanates and carbamates. It can also produce very wide range of sophisticated phosgene derivatives such as OCA monomer, acyl chloride and the NCA/UNCA anhydrides needed for polymers and for synthesis.

The company is also able to undertake custom phosgene synthesis based on client specific process and analytical development, QA management and safety/risk assessment programs.

About phosgene

Phosgene (carbonyl dichloride) is a chemical compound with the formula COCl2. Basic production is achieved by passing purified carbon monoxide and chlorine gas through a bed of porous activated carbon, which forms the catalyst for an exothermic reaction in a cooled reactor. This forms a colorless gas that became infamous as one of the first chemical weapons. This highly toxic character means extraordinary precautions to keep phosgene contained.

Despite its dark history, phosgene chemistry delivers essential benefits for activated amino acids, peptide synthesis and many other areas of pharmaceutical and fine chemical production. Phosgene-derived intermediates are used in manufacture of pharmaceutical active ingredients as well as specialty plastics, coatings, and agricultural protection chemicals.

Conventional derivatives

ISOCHEM offers a number of conventional phosgene derivatives requiring one or two synthesis steps, in volumes ranging from scores of metric tonnes to several hundred tonnes, using either batch or continuous production processes. These include chloroformates andchloroethyl chloroformates, acid chlorides, isocyanates, carbamates alpha-chloro carbonates, carbamoyl, chloro-heterocycles and ureas.

Conventional derivative manufacture is based on ISOCHEM’s long experience with phosgene and phosgene derivative chemistry, processes and safety.

Sophisticated phosgene derivatives

ISOCHEM can also produce a very wide range of sophisticated phosgene derivatives including O-carboxyanhydride (OCA) monomer, acyl chlorides and that are functional intermediates in the production of peptides, copolymers and hydroxyacyl derivatives.

NCA production

One of the most important functional intermediates is N-carboxy anhydride (NCA), which is an amino acid derivative that forms a basic building block in multistep synthesis or polymer precursors.

ISOCHEM has developed renowned expertise in phosgene-derived NCAs, using powerful processes leveraged from patented innovations to deliver NCA with the degree of purity required for polymerization or chemical synthesis.

The company’s industrial expertise in this type of chemical process enables production runs ranging from several tons to several dozen tons per year.

UNCA production

ISOCHEM also produces urethane N-carboxy anhydrides (UNCAs) for solution-phase short peptide synthesis processes. UNCA production can be carried out in batches of up to 100 kilograms.

Custom phosgene synthesis

Along with its routine phosgene-based products, ISOCHEM can provide customer-exclusive process and analytical development, safety and risk analysis, QA management and environmental hazard (EHS) assessment.

GMP or non-GMP phosgene chemistry

Three of ISOCHEM’s plants are equipped to carry out phosgenation and phosgene chemistry to GMP or non-GMP standards, covering the broad range of market needs.

Vert le Petit plant, near Paris, is dedicated to pharmaceutical phosgene derivatives. Here, development, pilot production and large-scale production are dedicated to GMP intermediates or active ingredients.
