Antibody optimization
For BioSiteHisto, antibody optimization is a process in which pretreatment methods, antibody dilution, incubation times, blocking and detection conditions are fine tuned for each antibody under controlled laboratory conditions, measuring values of specificity, sensitivity and stability.
A key success factor is to select correct tissues to express antigens that are appropriately fixed and processed for the specified primary antibody. This is complex because even weakly expressing tissues are relevant, since they offer important information about sensitivity, specificity and signal to noise ratio.
Optimization of antibodies should always be performed with more than one antibody against the same tissue antigen to get results for comparison. Different pretreatment methods are used to fine tune antibody concentration and incubation times. This also helps avoiding one of the biggest challenges in successful optimization, the variations in staining results produced within a laboratory or between different laboratories.
Optibodies
BioSiteHisto-developed Optibodies™ are a series of mouse monoclonal antibodies, specified for the most clinically and diagnostically important markers.
Optibodies are fine-tuned to current clinical needs of today`s laboratory and are also tested against NordiQC quality control standards for control tissues and staining criteria.
The key criterion for each Optibody™ is specificity and high affinity towards its target epitopes. The antibody is also tailored to be flexible and reliable in use, with high consistency between lots.
