Overview

Nordic BioSite offers a research-based immunohistochemistry (IHC) service through its Finnish-based cGLP laboratory, BioSiteHisto.

BioSiteHisto provides high-quality routine and advanced immunohistochemistry (IHC) with progressive immunohistochemical techniques for human and animal studies. Nordic BioSite offers high quality primary antibodies, along with a wide range of related buffers, reagents, chromogens and other IHC supplies.

IHC Background

Since American pathologist, Dr. Albert Coons, first developed immunofluorescence method in the early 1940s, including the Nakane & Pierce immunoperoxidase method in the late 1960s, immunohistochemistry (IHC) has provided the leading method for detecting proteins and other antigens in tissue section by exploiting the principle of antibodies binding specifically to antigens in biological tissues.

Immunohistochemical staining provides a powerful research tool for diagnosis of abnormal cells such as those found in cancerous tumors and is widely used in basic research to understand the distribution and localization of biomarkers and differentially expressed proteins in different parts of a biological tissue. Novel next generation IHC provides important tools for mutation specific diagnostic.

Standardization and optimization of the IHC processes are the corner stone for effective and trouble-free IHC processes. Optimization and standardization of the pre-analytical and analytical phases of the staining procedure minimize risks for false negative and false positive interpretations during the post-analytical phase.

BioSiteHisto IHC services

BioSiteHisto has wide knowledge from the field of immunohistochemistry, which ensures high-quality routine and advanced IHC processes that feature advanced Immunohistochemical techniques for human and animal studies. These include highly sensitive polymer-based immunoperoxidase staining, which offers high signal to noise ratio. In addition, alkaline phospathase IHC, as well as double and multi-stainings and fluorochromogenic combinations are part of the services. BioSiteHisto chromogenic immunostainings can be performed using permanent chromogens as DAB, permanent red and permanent green.

Optimized protocols

BioSiteHisto has also optimized a wide selection of antibodies for human, xenograft and different animal tissues, using multi-tissue sections. Tailored optimization processes and custom made antibody performance testing are part of the laboratory service.

Advanced cGLP processes

BioSiteHisto’s excellent results in external quality assessments of IHC are based on Good Laboratory Practice compliant (cGLP) immunohistochemical processes. The BioSiteHisto laboratory uses the Thermo Fisher Scientific Lab Vision™ Autostainer immunostainer system, which is open system and it allows a wide range of immunoreagents or even customer’s own reagents, if needed. Two instruments ensure the high-capacity and short turnaround times.

HRP Immunoperoxidase staining

Immunoperoxidase staining with HRP (horse radish peroxidase) enzyme and DAB visualization enhances possibilities to study target antigens from tissue sections (Paraffin section & frozen sections). This classical method with nice hue of the hematoxylin offers great sensitivity and contrast with DAB.

Alkalinephosphatase immunostaining

In special cases, where pigmentation and pigmented cells are problematic and pigmentation interferes with interpretation of DAB visualized slides, alkalinephosphatase based immunostaining can offer extra value. Red chromogen (Permanent red) signal is easy to demerge from pigmented cells (e.g. from skin tissue and melanoma diagnostic). Permanent red offers also great fluorescence value, making it possible to interpret in bright field and dark field microscopy.
